Review of the reproductive physiology of the scallop, Pecten maximus, applicable to intensive aquaculture ArchiMer

This paper presents a summary of information on reproduction biology in the scallop, Pecten maximus, which is required for the successful reseeding and intensive aquaculture programs developed in France since 1982. Data are presented on the following subjects: environmental factors that sustain gametogenic activity and the time of year this activity occurs; biochemical changes that are associated with gametogenesis; the consistency of egg quality; and development of a simple test to predict egg quality.

Effects of dissolved mercury on embryogenesis, survival, growth and metamorphosis of Crassostrea gigas oyster larvae ArchiMer

The effects of mercury (Hg) concentrations ranging from 0 (control) to 1024 µg 1-1 upon embryogenesis, survival, growth and metamorphosis of Crassostrea gigas (Thunberg) oyster larvae were investigated. Embryogenesis was abnormal in 50 % of the individuals at 11 µg 1-1 . The 48 h LD50 for D·shaped, umbonate and pediveliger larvae were 33, 115 and 200 µg 1-1 respectively. The increase in LD50 was partially explained by the larval weight increase, although weight-specific tolerance to Hg was higher in smaller larvae. Growth, the most sensitive physiological process studied, was significantly retarded at 4 µg 1-1. Effects of dissolved mercury on embryogenesis, survival and growth of Mytilus galloprovincialis mussel larvae ArchiMer

The effects of Hg concentrations ranging from 0 (control) to 1024 mu g l(-1) upon embryogenesis, survival and growth of Mytilus galloprovincialis mussel larvae were investigated. Embryogenesis was abnormal in 50% of the individuals at 10 mu g l(-1). The 48 h LC(50) for D-shaped, early umbonate, late umbonate and eyed larvae were 51, 164, 322 and 383 pg l(-1) respectively. The LC(50) was an allometric function of ash-free dry weight with exponent b = 0.60. Larval growth was significantly reduced after 6 d exposure to 4 mu g Hg l(-1) and after 10 d exposure to 2 mu g l(-1). Embryonic external morphogenesis of Rhammatocerus conspersus (Bruner) (Orthoptera: Acrididae: Gomphocerinae) and determination of the diapausing embryonic stage Anais da SEB

The grasshopper Rhammatocerus conspersus (Bruner) (Orthoptera: Acrididae: Gomphocerinae) is an occasional pest in pasturelands of Rio Grande do Sul State. It is a univoltine species with an embryonic diapause. Nymphal and adult stages occur during the warmer months (November-March). Eggs were dissected periodically for characterization of embryonic external morphogenesis in 1994 and 1995. Ten embryonic stages were illustrated. Two diapausing stages were verified in R. conspersus: one at 25% and another at 50% of total embryonic development.

Detection of equine herpesvirus 1 DNA in a single embryo and in horse semen by polymerase chain reaction Arq. Bras. Med. Vet. Zootec.

The genome of one equine embryo and three equine semen specimens collected from a Brazilian farm were tested by polymerase chain reaction (PCR) for the presence of EHV-1 and EHV-4-specific timidine kinase (TK) sequences. The PCR detected specific EHV-1 TK gene sequences in all samples tested. The peripheral blood leukocytes (PBL) of the embryo donor mare also was amplified by EHV-1 TK primers. Infectious virus was not recovered from any specimens. The animals did not show any clinical signal of EHV-1 or EHV-4 infections. EHV-4 was not detected in the studied specimens. The results indicate that PCR was more sensitive than virus isolation in cell culture for detecting EHV-1 in semen of carrier horses.

Cryopreservation of mouse morulae in glycerol, sucrose and honeybee royal jelly Braz. J. Vet. Res. Anim. Sci.

Compacted mouse morulae were frozen at 0.3ºC/min. or 0.5ºC/min. from -6ºC to -24ºC or -32ºC in 10% of glycerol plus different sucrose concentrations with or without 0.1% of honeybee royal jelly. Embryos were thawed in water bath at 22ºC for 20 seconds and cryoprotectant dilution was done in three steps. Embryos were cultured in Whitten’s medium for 24, 48 and 72 hours at 37ºC, 5% of CO2 and 100% of humidity. The in vitro development ranged from 56.6% to 100% after 72 hours. Expanded blastocysts were transferred to pseudopregnant recipients on the third day of the estrous cycle. The in vitro and in vivo development of mouse morulae after cryopreservation through different methods was examined. The slow-freezing involved an equilibration in 1.5M ethylene glycol (EG) and cooled at 0.5; 0.7; 1.0 or 1.2ºC/minute. The vitrification involved a 3 minutes equilibration in 20% EG and 60 seconds in solution containing 40% EG, 18% ficoll and 10.26% sucrose. The quick-freezing involved an equilibration in 3M EG + 0.3M sucrose for 5 minutes and 2 minutes in nitrogen vapor. Genetic components affecting embryonic developmental time of Drosophila melanogaster Genet. Mol. Biol.

The developmental time of the embryonic stage of Drosophila melanogaster was 21.66% faster and 14.75% slower than controls in populations selected for fast and slow developmental speed, respectively. The genetic model with two main loci with dominant and additive effect added to maternal effect and their relevant interactions can explain 96% of the phenotypic variability in the embryonic developmental time according to 14 crossing progenies involving fast and slow flies.

Species-specific differences in diatom-induced anomalies in calanoid copepod embryogenesis: are they linked to the stock-piled antioxidants? IBSS Repository

Egg production and viability of co-occurring in nature copepods, Calanus helgolandicus and Calanoides carinatus, fed diatom, Thalassiosira rotula, and then switched to dinoflagellate Prorocentrum minimum, were compared in experimental conditions. Egg production of both species on diatom diet was similar (up to 17-22 eggs.female-1.d-1) but no viable nauplii were observed. A study was carried out to evaluate the effect of broiler breeder age on the development of the digestive tract of embryos and chicks. Fertilized eggs Cobb from 30 and 60 week-old broiler breeder was utilized in this experiment. The results showed that eggs from older (60 weeks of age) broiler breeders were heavier (p = 0.001) than those from younger (30 weeks of age) broiler breeder. Embriogenesis and larval ontogeny of the "piau-gordura", Leporinus piau (Fowler) (Pisces, Anostomidae) after induced spawning Zoologia

Mature males and females of Leporinus piau (Fowler, 1941) were induced to reproduce through a hypophysation process. Extrusion occurred 12 h or 312 hours-degree after the hypophysation, at a water temperature of 26°C. Fertilized eggs were maintained in incubators at 24°C. Embryonic development, was evaluated using every 10 minutes, fresh egg samples which were analyzed under stereoscopic microscope. The larvae were collected at 24-hour intervals for seven days after hatching, fixed in Bouin's fluid and were submitted to routine histological techniques. Influence of environmental conditions on early development of the hydrothermal vent polychaete Alvinella pompejana ArchiMer

Dispersal and colonisation processes at deep-sea vents are still not fully understood, essentially because early life stages of vent species remain unknown. The polychaete worm Alvinella pompejana forms colonies on chimney walls at East Pacific Rise vent sites where the temperature can frequently exceed 20 degrees C. In vitro studies in pressure vessels showed that the early embryos tolerate temperatures in a lower range (10-14 degrees C), suggesting that they would have to escape the colony to develop. The effect of cryopreservation in IFN-tau, from bovine embryos produced in vitro was evaluated. Two treated groups (G1= fresh bovine embryos, n=59 and G2= freezed embryos, n=84) were used to study the effect of cryopreservation on IFN-tau secretion. After reaching the blastocyst phase, the embryos were kept on individual culture for additional period of 7 days. On days 3 and 7 after the beginning of embryos cultivation, samples of the media culture were taken for IFN-tau secretion titration. Oxidative modifications of cell components due to the action of reactive oxygen species (ROS) is one of the most potentially damaging processes for proper cell function. However, in the last few years it has been observed that ROS participate in physiological processes. The aim of this work was to determine ROS generation during in vitro production of bovine embryos. Cumulus-oocyte complexes were recovered by aspiration of antral follicles from ovaries obtained from slaughtered cows and cultured in medium 199 for 22 h at 39ºC in 5% CO2: 95% humidified air. Eggs of the Southern Brazilian flounder Paralichthys orbignyanus were obtained through spawning induction and reared in laboratory. The eggs released were free-floating or pelagic. Other important characteristics were: spherical shape, smooth chorion, narrow perivitelline space, and a single oil globule. Egg and oil globule diameter were about 0.792 ± 0.008 mm and 0.114 ± 0.003 mm, respectively. The blastodisc was observed approximately 1 h after fertilization and followed by blastodisc cleavage. Mango can be propagated by seeds or by grafting. For commercial purpose, grafting is the most appropriate method because it maintains the genetic characters from the propagated variety. To obtain grafted mango it is important to use polyembryonic varieties as rootstock since they produce a zygotic and many nucellar plantlets. The nucellar plantlets maintain the genetics of the mother-plant thus, are preferred for grafting since they supposedly give more uniformity to the orchard. We found that the sex-ratio of an amphigenic strain of Sciara ocellaris varied widely from progenies with few males to progenies containing a larger proportion of males, with single-sex progenies being rare. The sex-ratio distributions were dependent on the temperature at which the stocks of flies were raised, with the sex-ratio distributions being symmetrical (i.e. about 50% males) at 18 °C and 20 °C while at the higher temperatures of 24 °C and 28 °C the distributions were skewed toward a high proportion of females with the mean proportion of males decreasing to about 30-37% per progeny. Standardized experimental protocols designed to study Pacific oyster (Crassostrea gigas) gamete quality have not been previously published. Gamete sampling variations and confounding effect of interacting factors result in large variations between replicates, decreasing the effects of the studied factors. This work aims at defining a standardized procedure for incubation of oyster embryos designed for experimental purposes. In a first phase, four experiments were developed to improve embryo sampling and handling. Light and electron microscopy study of late embryonic development in the land snail Limicolaria flammea (Müller) (Pulmonata, Achatinidae) Zoologia

The late stages of embryogenesis in the achatinid land snail Limicolariaflammea (Müller, 1774) were described using light and electron microscopy. Embryos at various stages of development were present in the eggs during the first hour after they were laid, from 4-cell blastulae to morulae and fairly advanced stages. The advanced embryo which was fully developed on the second day bears a long cephalic sac, first to be developed, attached to the embryo and a podocyst which is attached to the foot of the embryo. Both of these structures are reduced in size as embryogenesis progresses until they finally disappear at about the 7th day after the egg was deposited.
